1 Detection Principle
Ractopamine (Rac) Rapid Detection Card applies the principle of competitive inhibition immunochromatography, ractopamine in the sample binds to the specific monoclonal antibody labeled with colloidal gold during the flow process, inhibits the binding of the antibody to the ractopamine conjugate on the NC membrane detection line (T line), resulting in a change in the color depth of the detection line (T line); According to the color development depth judgment result of the T line and the C line.

4, sample pretreatment method
(1) Take an appropriate amount of tissue samples that have been peeled and degreased in a homogenizer;
(2) Weigh 5.0 g0.05g homogenized tissue samples into a 10 mL centrifuge tube, buckle the tube cover, not too tight;
(3) Put it in a water bath at 80 ° C for 10 minutes and then take it out, cool it to room temperature, and absorb a clear liquid for inspection;
* If the liquid to be tested is viscous, it can be diluted 1:1 with clenbuterol dilution (3 drops of dilution + 3 drops of sample) for inspection.
* If the liquid has obvious yellow turbidity, centrifugation is required.
5, operation steps
(1) Take out the detection card from the original packaging bag, place it horizontally on the front of the observer, and use it as soon as possible after opening;
(2) Absorb 80 μL with a pipette (or 3 drops with a disposable straw) Before processing the liquid to be tested, add it vertically and slowly dropwise to the detection card and the sample hole (S);
(3) Start timing when the liquid flows, and the reaction takes 5 to 6 minutes. According to the schematic determination result, the other time is determined to be invalid.
6, the result judgment
Negative (-): T-line (detection line, near the end of the sample hole) color development is deeper than or equal to C-line (control line) color development, which means that the concentration of the substance to be tested in the sample is lower than the detection limit or no residue of the substance to be tested.
Positive (+): The color development of the T-line is lower than that of the line, or the color development of the T-line is not, which means that the concentration of the substance to be tested in the sample is higher than the detection limit; the shallower the T-line is than the C-line, indicating that the concentration of the substance to be tested in the sample is higher.
Invalid: The control line (C) does not appear a purplish-red strip, indicating that the operation process is incorrect or the detection card has failed. In this case, the instructions should be read carefully again and re-tested with a new detection card. If the problem still exists, do immediately stop using this batch number of products and contact the supplier.
