1, method principle
Nitrite and naphthalene hydrochloride ethylenediamine reaction under certain conditions to produce purple products, the color of the solution is proportional to the content of nitrite in the sample, the use of supporting food safety detector can determine the content of nitrite in the sample.
Second, the scope of application
This kit is suitable for meat products, vegetables, milk powder, salt and beverages and other food in the determination of nitrite content.
Three, you need to bring your own tools
scissors/cooking machine, balance, vortex meter, purified water, pipette (1-5mL), filter paper, timer, 10mL centrifuge tube
Four, sample measurement
1, weighed 1 gram of the minced sample (liquid sample directly absorbed 1mL) in 10mL centrifuge tube, add 4 mL of purified water, vortex oscillation for 2 minutes; continue to add 1mL reagent A, 1mL reagent B, with purified water bandwidth evaluation to 10mL, mixing, filtering, filtrate as sample treatment solution.
2, according to the number of samples tested, take an appropriate amount of 10mL centrifuge tube, mark on the control tube and sample tube.
Control tube: 1mL sample treatment solution, 0.3mL reagent C, 5mL purified water, 0.3mL purified water, mixed well into a cuvette;
Sample tube: 1mL sample treatment solution, 0.3mL reagent C, 5mL purified water, 0.3mL reagent D, mixed well into a cuvette;
3, refer to the method of use of the food safety detector, determine the control solution and sample solution respectively, and the detection result is automatically calculated by the detector.
Five, detection limit
The detection limit of nitrite in this kit is 1.0 mg/kg.
6. Precautions
1. If the color is very dark after color development and there is precipitation or soon fades into light yellow, it indicates that the nitrite content in the sample is very high. The sample treatment solution in the sample determination step 1 must be diluted with purified water by a certain multiple and then measured again. The nitrite content in the sample = the dilution multiple of the content determined by the instrument;
cases: The sample with deep color is diluted 10 times and then measured again. Then 0.5 mL of the sample treatment solution in the sample determination step 1 (the volume to be diluted can be selected according to the amount of the remaining sample treatment solution), 4.5 mL of purified water (the dilution multiple is 10) is added, and after mixing, it is used as a new sample treatment solution for subsequent testing. The final sample nitrite content = the content determined by the instrument 10.
2, when using this kit to determine samples, should be carried out under ventilated conditions;
3, all reagents should avoid contact with the skin, such as inadvertent contact, should immediately rinse with plenty of water.
